Volatile organic compounds (VOCs) are secondary metabolites acting as a language for the communication of plants with the environment. In orange fruits, the monoterpene D-limonene accumulates at very high levels in oil glands from the peel. Drastic down-regulation of D-limonene synthase gene expression in the peel of transgenic oranges harboring a D-limonene synthase transgene in antisense (AS) configuration altered the monoterpene profile in oil glands, mainly resulting in reduced accumulation of D-limonene. This led to fruit resistance against Penicillium digitatum (Pd), Xanthomonas citri subsp. citri (Xcc) and other specialized pathogens. Here, we analyze resistance to pathogens in independent AS and empty vector (EV) lines, which have low, medium or high D-limonene concentrations and show that the level of resistance is inversely related to the accumulation of D-limonene in orange peels, thus explaining the need of high D-limonene accumulation in mature oranges in nature for the efficient attraction of specialized microorganism frugivores.  相似文献

Summary The wide range of functional activities of circulating and sessile insect hemocytes expresses itself in highly specialized cytological terms. Electron microscopic studies carried out in five species of normal and experimentally manipulated cockroaches, in conjunction with light microscopic information, reveal a broad spectrum of structural organization and an apparent capacity for cellular modulation. In addition to conventional organelles, these hemocytes contain a class of unusual cytoplasmic inclusion bodies which seem to undergo striking transformations in response to specific functional demands. A variety of transitional forms attests to the existence of close links between tubule containing (type 1), electron dense (type 2), and large globular (type 3) inclusion bodies, and reveals the derivation of yet another special (lamellated, fusiform) inclusion from type 2 bodies. Confluence of the type 3 configurations into still larger lacunae may precede the release of their contents into the hemolymph, a process whose major effect seems to be the initiation of the clotting process.Another important activity of hemocytes concerns the programmed reorganization of the stromal framework of the various organs. The dominant feature of blood cells engaged in the deposition of connective tissue are greatly distended cisternae of the rough endoplasmic reticulum and accumulations of banded fibrils at the interface of cytoplasm and extracellular space. The engulfment of discarded stromal material can be visualized in fortuitous sections representing steps in its incorporation by hemocytes. Ultrastructural correlates of the breakdown of these and other disintegrating or noxious elements by certain hemocytes are prominent digestive vacuoles with heterogeneous contents and reaction products of hydrolytic enzymes. The capacity for the uptake of small particles by micropinocytosis is demonstrated by the localization of horseradish peroxidase activity at the cellular surface and within cytoplasmic vesicles.The diversity of structural appearances reflects a division of labor, while the many transitional features of hemocyte morphology favor the concept of functional flexibility of one basic cell type rather than a strict classification into distinctly separate cellular types.Supported by research grants NB-05219, NB-00840, and 5 P01 NS-07512 from the U.S.P.H.S.I am greatly indebted to Dr. Joseph Osinchak, City University, New York, who has generously permitted me to include some of his micrographs in the present study. RHO GTPases are key regulators of the cytoskeletal architecture, which impact a broad range of biological processes in malignant cells including motility, invasion, and metastasis, thereby affecting tumor progression. One of the constraints during cell migration is the diameter of the pores through which cells pass. In this respect, the size and shape of the nucleus pose a major limitation. Therefore, enhanced nuclear plasticity can promote cell migration. Nuclear morphology is determined in part through the cytoskeleton, which connects to the nucleoskeleton through the Linker of Nucleoskeleton and Cytoskeleton (LINC) complex. Here, we unravel the role of RAC1 as an orchestrator of nuclear morphology in melanoma cells. We demonstrate that activated RAC1 promotes nuclear alterations through its effector PAK1 and the tubulin cytoskeleton, thereby enhancing migration and intravasation of melanoma cells. Disruption of the LINC complex prevented RAC1-induced nuclear alterations and the invasive properties of melanoma cells. Thus, RAC1 induces nuclear morphology alterations through microtubules and the LINC complex to promote an invasive phenotype in melanoma cells.  相似文献

Blocking the PD-1/PD-L1 pathway has emerged as a potential therapy to restore impaired immune responses in human immunodeficiency virus (HIV)-infected individuals. Most reports have studied the impact of the PD-L1 blockade on effector cells and neglected possible effects on regulatory T cells (Treg cells), which play an essential role in balancing immunopathology and antiviral effector responses. The aim of this study was to define the consequences of ex vivo PD-L1 blockade on Treg cells from HIV-infected individuals. We observed that HIV infection led to an increase in PD-1+ and PD-L1+ Treg cells. This upregulation correlated with disease progression and decreased under antiretroviral treatment. Treg cells from viremic individuals had a particularly high PD-1 expression and impaired proliferative capacity in comparison with Treg cells from individuals under antiretroviral treatment. PD-L1 blockade restored the proliferative capacity of Treg cells from viremic individuals but had no effect on its suppressive capacity. Moreover, it increased the viral production in cell cultures from viremic individuals. This increase in viral production correlated with an increase in Treg cell percentage and a reduction in the CD4/Treg and CD8/Treg cell ratios. In contrast to the effect of the PD-L1 blockade on Treg cells from viremic individuals, we did not observe a significant effect on the proliferative capacity of Treg cells from individuals in whom viremia was controlled (either spontaneously or by antiretroviral treatment). However, PD-L1 blockade resulted in an increased proliferative capacity of HIV-specific-CD8 T cells in all subjects. Taken together, our findings suggest that manipulating PD-L1 in vivo can be expected to influence the net gain of effector function depending on the subject’s plasma viremia.  相似文献

High‐resolution X‐ray computed tomographic scans were used to examine pterygoid sinus morphology within extant porpoise species and one delphinid (Tursiops truncatus), in order to consider: 1) intraspecific and interspecific variation among the studied species; 2) the most parsimonious sequence of character acquisition; and 3) the potential functional roles of the preorbital lobes of the sinuses in sound reflection. Scans revealed that the pterygoid/palatine regions are mediolaterally broader in the earliest diverging phocoenid (Neophocaena phocaenoides) and Tursiops truncatus than the dorsoventrally elongated sinuses observed in other species. Rostrocaudal lengths of the sphenoidal regions of the sinuses in all individuals studied are proportionally similar, indicating conservatism in this region across species. The neonate Phocoena phocoena has shorter preorbital lobes than adults, but they are still proportionally longer than Neophocaena phocaenoides and Phocoena spinipinnis. The preorbital lobes broaden mediolaterally to varying degrees across species; in particular, Phocoenoides dalli has the largest dorsal and lateral expansion of this region. Assuming the highest pulse frequency produced by porpoises is 150 kHz, all regions of the preorbital lobes are thick enough to reflect the wavelengths produced. In addition, the neonate preorbital lobes are not as elongated as they are in adults, and the dorsal third of this region may not reflect sound to the same extent. This study reinforces the importance of using nondestructive methods to quantify variation in endocranial anatomy and the value of CT data for recovering phylogenetically useful information, as well as functional roles sinuses play in concert with the soft tissue head anatomy for biosonar. Biodiversity conservation strategies that overlook the interests of local people are prone to create conflicts. The ecosystem service approach holds potential for more comprehensively integrating the social dimension into decision-making in protected areas, but its implementation in conservation policies is still in its infancy. This research assesses the extent to which ecosystem services have been implemented in conservation strategies in protected areas. The study was conducted in two outstanding Spanish protected areas, covering a wetland (Doñana Natural and National Parks) and a Mediterranean mountain system (Sierra Nevada Natural and National Parks). Data were collected from deliberative workshops with managers and researchers, face-to-face surveys with users and a review of management plans. We found that, beyond intrinsic values of ecosystems and biodiversity, these areas provide multiple ecosystem services that deserve further attention to ensure their sustained delivery. Our research shows that environmental managers and researchers have different perceptions and priorities regarding ecosystem services management compared with ecosystem service users. Environmental managers and researchers in both protected areas perceived that human-nature relationships and ecosystem services are already widely included in management plans, if often not explicitly. We found that different ecosystem service categories receive uneven attention in management plans. These contained measures to manage provisioning and cultural services whereas measures for managing regulating services were perceived to be largely absent. We conclude by summarizing insights on how the ecosystem service approach may enhance the consideration of social interests in the management of management protected areas.  相似文献
